    phenothiazines: (fe?no-thi'a-zenz) A class of major tranquilizers used to treat psychotic illnesses such as schizophrenia. They have neuroleptic and antiemetic effects. Among the most commonly used agents in this class are chlorpromazine, haloperidol, prochlorperazine, and thioridazine. Side effects of these drugs include dystonic reactions. The antischizophrenic activity of phenothiazine drugs and their tendency to elicit extrapyramidal symptoms are thought to involve blockade of synaptic dopamine receptors in the brain. Space filling molecular models show how favorable Van der Waal's interactions between the side chain amino of phenothiazines and the 2-substituent on ring A can.

Phenothiazine was initially synthesized in by Bernthsen. It was the basis for the development of other drugs including the phenothiazine class of antipsychotics or neuroleptics.
